Atomic force microscope (AFM) is used to study restriction endonuclease digestion of plasmid DNA. pWRr plasmid DNA is digested by Hind III, and the specific and the nonspecific binding of the restriction endonuclease are imaged, and the biological function of the enzyme binding to nonspecific sites is discussed. In addition, it is found that nonspecific binding of Hind III could not induce the DNA characteristic bending angle.
